The Oranje have lost Feyenoord midfielder Clasie to a thigh problem and Aston Villa defender Vlaar with a calf strain.
The two players will also miss out on the Euro 2016 qualifier against the Czech Republic.
They called up Gregory van der Wiel and Virgil van Dijk as replacements.
Coach Guus Hiddink had already sent home Arjen Robben, Klaas-Jan Huntelaar and Rafael van der Vaart.
