Destro had joined the Giallorossi on loan worth €11.5m last summer, though his campaign was blighted by injuries and poor form.
The 22-year-old managed six goals in 21 Serie A appearances, plus five in five Coppa Italia games.
Roma released a statement confirming they exercised their option to buy Destro outright for a further €4.5m, taking his total transfer value to €16m.
The same statement also outlined the financial details of some other transfers completed this week.
Roma paid €2m to buy out Pescara’s half of midfielder Gianluca Caprari, plus €1.5m to take Giammario Piscitella and €2.5m for Valerio Verre, who were both in co-ownership deals with Genoa.
In turn, Genoa paid €3m to buy out Roma’s half of Panagiotis Tachtsidis.
